CLINICAL IMPLICATIONS: Effects of treatment position on drug delivery and therapeutic outcomes should be considered when prescribing nebulized bronchodilators.

Protesters In Haiti's Capital Demand Aristide's Return Nasdaq February 9, 2007 PORT-AU-PRINCE, Haiti AP ; --Hundreds of supporters of ousted former president Jean-Bertrand Aristide protested outside the U.N. peacekeeping mission Wednesday, demanding the exiled leader's return and accusing blue-helmeted troops of killing civilians. Protesters shouted "Down with the U.N.!" as they marched from a downtown slum and gathered outside the U.N. mission's hilltop headquarters, where they shouted at U.N. soldiers and Haitian police guarding the fortified complex. "We want the international community to hear our voice when we say we want Aristide back, " Jacques Bloncourt, 48, said as he held aloft a photograph of the former leader, who is currently in exile in South Africa. Aristide, a former slum priest turned president, fled the Caribbean nation in February 2004 amid a three-week rebel uprising. He later accused the U.S. of kidnapping him in a coup - a charge Washington denies. Many of the protesters came from the seaside slum of Cite Soleil, an Aristide stronghold where armed gangs and U.N. peacekeepers clash almost daily. The protesters said civilians are often killed in gunbattles started by peacekeepers and accused President Rene Preval of allowing it to happen. "President Preval lets them kill us, " Cite Soleil resident Fredrick Dubois said. The 9, 000-strong U.N. peacekeeping force arrived in July 2004 to quell violence after Aristide's ouster but has struggled to root out gangs based in Port-au-Prince's dense slums. Core trainers for the transition team must be carefully chosen in order to provide comprehensive training, set performance criteria, and establish realistic time frames for completion of training. Once the transition period has begun, progress must be monitored closely by lead managers of the respective companies. Daily, weekly, and monthly goals must be established and progress reviewed regularly. Feedback on performance needs to be solicited from investigators, department managers, and professional staff through surveys and face-to-face discussions. At our institution, the transition was completed within the designated time frame with minimal impact on in-house research. Animal care, health, welfare, and compliance were never jeopardized. If there are any displaced employees, they should be treated professionally and with compassion. By maintaining clear objectives and time frames as well as open communication with feedback at multiple levels, a smooth transition can be achieved. The key is active participation of both parties in the transition process coupled with treating all employees with respect and dignity during the process. PS30 Persistent Colonization of Cats with Helicobacter pylori: A Possible Zoonosis in Vivaria MI Esteves, * RP Marini, S Perkins, NS Taylor, L Handt1, JG Fox Massachusetts Institute of Technology, Cambridge, MA 02139, 1Merk Research Laboratories, West Point, PA 19486 Helicobacter pylori was isolated from one colony of commercially reared cats in 1994. To ascertain prevalence and whether persistent natural infection occurred in these animals, cats of different ages, including 6 cats undergoing serial endoscopic biopsies over 1.5 years, were screened for the presence of H. pylori in salivary secretions, gastric juice, gastric tissue, and feces. Group 1 comprised 12 cats 1year old; of these, H. pylori was cultured from stomach biopsies in 9 12 75% ; animals. Group 2 included 26 cats aged one to two years old; 26 100% ; had positive H. pylori cultures of gastric mucosa; 11 12 91% ; and 6 of 12 50% ; were culture positive from the gastric and salivary secretions, respectively. Group 3 consisted of 4 animals 5-6 years old; these cats had 3 4 75% ; positive cultures from gastric fluid. The fecal cultures of 15 cats examined were negative for H. pylori but 4 5 80% ; of group 2 cats were positive by PCR analyses of feces. Six cats selected from group 2, examined at 8, 11, 22, and 27 months of age all demonstrated persistent gastric colonization with H. pylori. ACTIONS Albuterol is primarily a beta-2 sympathomimetic and as such produces bronchodilation. Because of its greater specificity for beta-2 adrenergic receptors it produces fewer cardiovascular side effects and more prolonged bronchodilation than isoproterenol. Onset is within 15 minutes; peaks in 60-90 minutes. Therapeutic effects may be active up to 5 hours. INDICATIONS Albuterol inhaler is indicated for relief of bronchospasm in patients with reversible obstructive airway disease including asthma. CONTRAINDICATIONS Albuterol is contraindicated in patients with a history of hypersensitivity. WARNINGS Use cautiously in patients with coronary artery disease, hypertension, hyperthyroidism, and diabetes. Epinephrine should not be used subsequent to a failure of the other. Administer cautiously to patients on MAO inhibitors or tricyclic anti-depressants. Beta-Blockers and Albuterol will inhibit each other. Polyamines are powerful modulators of both growth and survival in mammalian cells. In this study, we investigated the possibility of attenuating the process of apoptosis in bone marrow stromal cells BMSCs ; , which comprise mesenchymal stem cells, by reducing the intracellular levels of polyamines. BMSCs were isolated from rat femurs and expanded for 12 days. At this time, BMSCs were CD34neg, CD45neg, and mostly CD90pos . BMSCs were grown for an additional 2 days in the presence of 1 mM -difluoromethylornithine DFMO ; , an inhibitor of ornithine decarboxylase, which reduced the content of both putrescine and spermidine by nearly 90%. DFMO treatment progressively slowed down BMSC proliferation, as determined by the 3- 4, 5-dimethylthiazol-2-yl ; -2, 5-diphenyltetrazoliumbromide MTT ; assay, without arresting their growth completely. The effect of polyamine depletion on caspase-3 activity was evaluated in BMSCs after treatment with 500 U ml tumor necrosis factor- TNF ; and 5 M MG132 , an inhibitor of proteasome. Caspase-3 activity increased linearly over a period of 24-hour stimulation p .01 ; , but this augmentation was blunted by 50% after DFMO administration p .05 ; . The effect of DFMO on TNF MG132 -induced upregulation of caspase-3 activity was reversed by the addition of 100 M putrescine, confirming that polyamines were really involved in the apoptotic process. Also, the number of apoptotic BMSCs after TNF MG132 treatment, as determined by terminal transferase-mediated dUTP nick endlabeling TUNEL ; assay, were threefold reduced after polyamine depletion p .05 ; . On the contrary, DFMO did not affect the MG132 -mediated increase in p53 abundance, nor its translocation to the nucleus.
